The Collaborative Study on the Genetics of Alcoholism: Overview PMC

A series of functional genomics studies examine the specific cellular and molecular mechanisms underlying AUD. This overview provides the framework for the development of COGA as a scientific resource in the past three decades, with individual reviews providing in‐depth descriptions of data on and discoveries from behavioral and clinical, brain function, genetic and functional genomics data. The value of COGA also resides in its data sharing policies, its efforts to communicate scientific findings to the broader community via a project website and its potential to nurture early career investigators and to generate independent research that has broadened the impact of gene‐brain‐behavior research into AUD. This review describes the genetic approaches and results from the family-based Collaborative Study on the genetics of alcoholism (COGA).

With current review, we aim to present the recent advances in genetic and molecular studies of AUDs. IDENTIFYING AN INTEGRATED APPROACH FOR AUD RESEARCH

For anxiety disorders, a prevalence of 22.2% has been reported among adults in Uganda [5]. Combined together, depression and anxiety disorders have been reported to affect approximately one in four persons in Uganda [5]. COGA researchers have also analyzed candidate genes—genes suspected to play a role in the development of alcoholism based on other studies. Some of these candidate genes encode components of various brain chemical systems that allow communication among nerve cells.

BRAIN FUNCTION

EEGs measure overall brain activity, whereas ERPs are brain waves elicited in response to specific stimuli (e.g., a light or sound). Analysis of such electrophysiological data may reveal a subset of genes that affect these quantitative, biological phenotypes related to alcoholism (Porjesz et al. 1998, 2002). One component of an ERP is a brain wave called P300, which typically occurs 300 milliseconds after a stimulus. Previous studies had found that a reduced amplitude of the P300 wave is a heritable phenotype that correlates with alcohol dependence and other psychiatric disorders (Porjesz et al. 1998).

Since common mental disorders are quite prevalent in the Uganda genome resource, there is opportunity to collect the phenotype data and link it with the existing genetics data to conduct genome-wide association studies for several mental disorders. The Uganda genome resource is also poised to grow in sample size and will include proteomics, metabolomics, and single-cell genomic studies which can advance functional genomics research for mental conditions in Uganda and Africa at large. Also, given the longitudinal design of the GPC, there is opportunity to track changes and understand the dynamics of mental disorders and their genetic underpinnings and can ultimately provide insights into causality and progression of mental disorders. These data continue to serve, not only as a platform for characterization of loci discovered in our own GWAS of behavioral and brain data but also for emerging signals from larger scale meta‐analytic GWAS of AUD. Another study investigating the heritability of assorted substance dependencies, including alcohol, tobacco, cannabis, and illicit drugs, used GCTA estimates to conclude that common SNPs contribute to at least 20% of the variance in substance dependence vulnerability (Palmer et al., 2015).
